MATERIALS AND METHODS Organism. M. smegmatis H-607 3 ; was grown routinely in 200-ml volumes of modified Sauton synthetic liquid medium 2 ; at 37 with aeration rotary shaking at 150 rpm ; . Under these conditions, the organism was markedly inhibited by as little as 0.2 Ag of ethambutol free base ; per ml. Growth was estimated turbidimetrically. Optical densities were measured at 600 nm in 18 150 mm tubes with a Coleman Junior spectrophotometer. An optical density of 0.2 was equivalent to about 0.3 mg dry weight ; of cells per ml. Ethambutol. Ethambutol dihydrochloride Myambu6ol ; was a gift from Lederle Laboratories, Pearl River, N.Y. The preparation of 14C-labeled ethambutol was described earlier 1 ; . This material demonstrated biological activity equivalent to that of unlabeled drug, and it had a specific activity of 5.12 gCi per mg 6.92 uCi per mg of free base ; . '4C-ethambutol uptake experiments. Overnight log-phase cultures were combined and adjusted to an optical density of 0.2 with fresh Sauton medium. Either 75- or 100-ml portions of adjusted suspension were placed in 500-ml Erlenmeyer flasks. "4C-ethambutol, metal salts, and spermidine were added from aqueous stock solutions. At selected time intervals during aerated incubation at 37 C, the cells contained in single flasks were collected on bacteriological filter membranes diameter, 47 mm; pore size, 0.8 Mm ; and washed three times with decreasing volumes of deionized water 30, 15, and 2 ml ; . To avoid corrections of data for filter-pad adsorption of 14C activity, each washed cell mass was scraped from the filter and transferred to a second tared membrane 2.54 cm in diameter. The samples were then dried, reweighed, and placed in scintillation vials containing 10-ml volumes of liquid scintillation counting cocktail. The cocktail consisted of 1 liter of toluene, 42 ml of Liquifluor Pilot Chemicals, Inc., Watertown, Mass. ; , and 40 g of Cab-o-sil Packard Instrument Co., Downers Grove, Ill. ; . After brief vigorous shaking of the vials, 14C activity was assayed with a Beckman LS-100C liquid scintillation system. The samples were counted either for 20 min or until 4, 000 counts had been collected, whichever came first. From 23 to 33 mg dry weight ; of cells was assayed in individual.

Assays. There is apparent specificity of this effect for the cholinergic PPI response. Adrenergic signaling through receptors was less sensitive than was cholinergic signaling through muscarinic presumably M1 ; receptors. Cholinergic binding affinities were in the same concentration range as the ED50 concentration, and the correlation of inhibitory potency with cholinergic affinity was the same as that with binding affinity though it should be noted that poor correlation was observed when [3H]quinuclidinyl benzilate was used as cholinergic ligand instead of [3H]Oxo-M Bowen et al. 1989b . However, several experiments ruled out direct antagonism of the cholinergic response i.e., a direct anticholinergic effect ; . The strongest evidence is that + ; -PENT inhibition of Oxo-M-stimulated PPI turnover could be observed under conditions where there was no occupation of [3H]Oxo-M binding sites. Also, based on the high binding affinity of Oxo-M to muscarinic receptors and the high concentration used to stimulate the response, there should be little occupation of muscarinic receptors by ligands under the normal conditions of the assay both ligands present together ; . The noncompetitive nature of the effect also argues against competitive antagonism at the receptor. However, the cholinergic receptor does appear to be involved in the mechanism of the effect. When intact synaptoneurosomes were pretreated with + ; -PENT, a marked decrease in the number of [3H]Oxo-M binding sites was observed. There was no change in the binding affinity. Thus, activation of receptors causes a down-regulation of cholinergic receptors. This down-regulation occurred in as little as 15 minutes. A mechanism involving decreased receptor number is consistent with the observed o-induced decrease in maximal.
